Correlations between cytotoxicity, biochemical effects, drug levels, and therapeutic effectiveness of daunomycin and adriamycin on Sarcoma 180 ascites in mice.

چکیده
The effects of adriamycin and daunomycin on mitotic activity, nucleic acid synthesis, and proliferative activity have been compared in Sarcoma 180 ascites in mice and related to the uptake and retention of the drugs by the ascites cells. The half-life of the drug in the cells after a single injection was 4 hr for daunomycin and 12 hr for adriamycin, while after repeated doses the uptake of the antibiotics was similar. The inhibition of mitotic activity and of thymidine incorporation into DNA was similar with daunomycin and adriamycin, while adriamycin inhib ited uridine incorporation into RNA to a greater extent. When given according to different schedules to Sarcoma 180-bearing mice, adriamycin appeared able to inhibit tumor growth to a greater extent than did daunomycin and to induce a greater number of tumor-free long-term survi vors. This effect has been observed also after a treatment schedule which results in similar intracellular levels of both antibiotics. The importance of mechanisms other than different cellular uptake of the drugs, operative in determin ing differences in the overall therapeutic efficacy in vivo, remains to be elucidated.

Effects of anthracyclines on oxygenated and hypoxic tumor cells.
The cytotoxic effects of anthracyclines and other chemotherapeutic agents were examined in normally aerated and hypoxic Sarcoma 180 and EMT6 tumor cells in vitro. Adriamycin, daunomycin, and mitomycin C were selectively toxic to hypoxic Sarcoma 180 cells. The augmented sensitivity was not the result of an increase in susceptibility of oxygen-deprived cells toward antitumor agents in general. 1,...
متن کاملEffects of quinidine and related compounds on cytotoxicity and cellular accumulation of vincristine and adriamycin in drug-resistant tumor cells.
Quinidine, which has antiarrhythmic activity, greatly enhanced the cytotoxicity of vincristine (VCR) in tumor cells and especially in VCR-resistant sublines of P388 leukemia (P388/VCR) and human myelogenous leukemia. A nontoxic concentration of quinidine increased VCR cytotoxicity in these resistant tumor cells about 50 to 80 times, and the drug in combination with VCR could completely reverse ...
متن کاملSome inhibitory effects of (--)-emetine on growth of Ehrlich ascites carcinoma.
(-)-Emetine has little or no effect on O(2) consumption of Ehrlich ascites-cell suspensions or on the viability of transplanted Ehrlich ascites-tumour cells exposed to, or incubated with, the drug in vitro before inoculation into new-host mice. (-)-Emetine administered as a single injection to mice bearing Ehrlich ascites-tumour cells slows the growth rate of the tumour. The subcutaneous or int...
متن کاملPhotoaffinity labeling of the Sarcoma 180 cell surface by daunomycin.
We have used photoaffinity labeling to investigate the distribution and function of daunomycin binding sites in Sarcoma 180 cells. When native daunomycin is irradiated at 366 or 488 nm in the presence of cells, the drug is irreversibly incorporated into cellular molecules. The cellular acceptor for the photoincorporation cannot be extracted by chloroform-methanol nor can it be degraded by DNase...
متن کاملProtein kinase C in adriamycin action and resistance in mouse sarcoma 180 cells.
Adriamycin has a wide variety of biological actions on susceptible cells, several of which may be integrally involved in cytotoxicity. In this paper, we present evidence that one of the alterations in cell function that occurs in the presence of Adriamycin is an elevation in the production of diacylglycerol. The effect is rapid, reaches a peak within 10 min of exposure of Sarcoma 180 cells to A...
